Background: The study aimed to assess the efficacy of various concentrations of sodium hypochlorite (NaOCl) solution in Enterococcus faecalis reduction in root canals. Methods: A total of 100 maxillary central incisors underwent chemomechanical preparation. Immersion in 5.25% NaOCl for 3 min removed smear layer followed by 1 mL of 17% ethylenediaminetetraacetic acid for 3 min. Phosphate-buffered saline solution was the final irrigant used. The samples were then sterilized. E. faecalis biofilms formed within the root canals were evaluated after 6 weeks. Samples were divided into five groups in terms of the antibacterial treatment: Group 1: 0.5% NaOCl solution; Group 2: 2.5% NaOCl solution; Group 3: 5.25% NaOCl solution; Group 4: Soaked in saline solution; and Group 5: Negative control. Microbial was quantified by method of colony-forming units followed by data analysis. Results: Higher concentrations of NaOCl demonstrated greater antimicrobial reduction as against 2.5% and 0.5% at P < 0.001. About 5.25% showed complete inhibition of microbial growth. The highest colonization was in the group irrigated with sterile saline solution. Conclusion: The present analysis leaned toward higher antiseptic concentrations to achieve optimum disinfection in the root canals.
